A comparative study on genomic relatedness of plasmodium falciparum among under-fives children in Morogoro.

Abstract
The aim of this study was to survey and determine the association between whole genome genetic differences among P. falciparum field isolates from under-five children. A total of 10 samples were used on surveying gene copy number variation from parasites isolated in children with different malaria severity and from different sites. Nineteen samples were used to survey total expression of parasites in erythrocytic stage isolated from children based on severity and space clustering. The differences were examined by using microarray technology and global positioning system. A total of 8787 oligonucleotides for desired segments were printed representing the whole genome of P. falciparum 3D7 strain, which was used as a reference as well. The results of hierarchical clustering (HCL) showed that the parasites were not genetically different from different malaria severity, sites or space clustering. However, when statistical analysis was conducted, 42 and 162 genes were significantly different (P=0.01) in copy number between parasites isolated from children with different malaria severity and sites, respectively. Furthermore, 57 and 61 genes were significantly different (P=0.01) on expression between parasites isolates based on severity and space clustering of malaria cases, respectively. Most of the genes associated with transmission and adaptation in vivo were not significantly different from any of the above cases, and this may suggest a common control approach. However similar studies with large sample sizes are required. In additional the genes that were significantly different have to be confirmed using other techniques such as quantitative real time Polymerase Chain Reaction (PCR).
